過去研究發現遷徙型的烏魚(M型)其磷酸葡萄糖異構酶之基因型為PGI-A^(100),而常駐型的烏魚、(R型)之基因型則為PGI-A句話,兩者在台灣海岸與河口地區分佈在不同的溫度範圍。本研究中針對三種不同基因型的烏魚、(pgi-A^(100/100),pgi-A^(100/135)與pgi-A^(135/135)),檢測極端溫度對熱休克蛋白(HSP70)表顯與磷酸葡萄糖異構酶活性的影響。低溫(5°C)4小時後,基因型pgi-A^(100/135)和pgi-A^(135/135)之烏魚幼魚其HSP70的水平高於基因型pgi-A^(100/100)的個體,且統計達到顯著(p < 0.05);高溫(35°C)4小時後,基因型pgi-A^(100/135)其HSP70的表現量上比pgi-A^(135/135)高,但沒有達到統計學意義(p > 0.05)。帶有pgi-A^(135)等位基因的烏魚幼魚接受溫度改變從20°C到35°C或5°C長達48小時之後,HSP70在0至8小時間有急遽上升的現象,但超過8小時35°C處理的個體其HSP70表現量會開始下降,而5°C處理的個體則仍維持高度的表現。

英文摘要

Two types of mullet, the migratory (M-type) type with PGI-A^(100) and the resident (R-type) type with PGI-A^(135), live in the environment of different temperatures around coastal areas and estuaries of Taiwan. In this study, we examined the effect of extreme temperatures on the expression of heat shock protein (HSP) 70 and the activities of phosphoglucose isomerase (PGI) of mullet larvae with 3 genotypes, pgi-A^(100/100), pgi-A^(100/135) and pgi-A^(135/135). Following 4 h exposure to 5°C, mullet larvae with pgi-A^(100/135) and pgi-A^(135/135) genotypes had significantly higher levels of HSP70 than individuals with pgi-A^(100/100) genotype (p < 0.05). However, the expression level of HSP70 of larvae with pgi-A^(100/135) genotype was not significant difference from that larvae with pgi-^(A100/135) genotype after exposure to 35°C for 4 h (p > 0.05). The expression level of HSP70 of larvae with the pgi-A^(135) allele exposed to temperatures changed from 20 to 35°C or from 20 to 5°C exponentially increased during 0~8 h exposure. The expression levels of the HSP70 of larvae with pgi-A^(135) genotype exposed to 5°C and 35°C from 8 h to 48 h showed a relative constant and a dramatic decrease, respectively.
