在培養的大腦皮質神細胞,乙醇保護神經細胞對抗NMDA興奮性毒性的效果仍有爭論。我們發現:在富含星狀膠原細胞的培養大腦皮質神經細胞(體內典型構造),事給與乙醇(10-100mM,30min)不能防止NMDA神經毒性(60-500Um,5-30min)所引起神經的死亡。評估神經的死亡,是採用相位差顯微鏡和培養液中乳酸去氫酵素的釋放。此結果建議:乙醇抑制NMDA神經毒性的能力,可能被乙醇對膠原細胞的作用所抵消。所以,乙醇可能對由NMDA引起的腦傷沒有好處。

英文摘要

The neuroprotective effect of ethanol on N-methyl-D-aspartate (NMDA) excitotoxicity in cortical cultures has been controversial. We show that application of ethanol (10-100 mM) on astrocyte-Rich cortical cultures, typical structure in vivo, dose not prevent the neuronal death induced by NMDA toxicity (60-500 uM, 5-30 min), assessed by phase-contrast microscopy and release of lactate dehydrogenase into medium. The results suggest that the ability of ethanol inhibiting NMDA neurotoxicity may be counter balanced by its actions on astrocytes. Therefore, ethanol may not be beneficial to NMDA mediated brain injury.
