We have developed a novel and effective method for production of transgenic goat by following a series of established procedure. We began with in vitro maturation of goat oocyte (IVM), followed by in vitro fertilization (IVF), and then followed by microinjection of the foreign gene (hFIX gene). We then identified the transgenic embryos, prior to their implantation, using nested PCR. Finally the integrated embryos were transferred into goat uterus. Using this new methodology, we transferred thirty-seven interated embryos into uteri of thirty-two goats. Nineteen of the goats became pregnant, giving a frequency of 60% (19/32) gestation. Out of these, three goats carried full-term pregnancy, and delivered four baby goats alive. The integrated frequency was 100% (4/4). The frequency of production transgenic goats by this new technique was 12.5% (4/32). It showed almost twice efficiency as that by the conventional “microinjection into pronuclei” method, with an average of 6.4% frequency. |
