Corneal-scleral rims were collected after corneal trasplantation as a source of cell culture materials. 0.8% Trypsin combined with 0.6% EDTA was used to disaggregate and harvest the endothelium; After cell culture in Earle's minimum essential medium with 10% Fetal calf serum for one month, the result are: In 12 specimens, one specimen was contaminated and discarded, 4 specimens were cultured to partial confluence successfully. 4 specimens had remained in stress form without proliferation. 3 cultures were failure and unable to survive. Analysis of the donor ages between successful group and failure group, the donor ages of successful group is not significant lower than that of failure gorup (p>0.05 z-test). As compare the corneal preservation period in organ culture, there are no significant difference within 3 groups and it may mean the duration of corneal preservation in organ culture do not influent the endothelial cell vitality.
