近年來隨著飲食及生活型態改變,國人慢性疾病發生率及盛行率逐年增加。野櫻莓含有豐富的酚酸及類黃酮,這些活性成分具有潛在的健康益處。故本研究探討野櫻莓之活性成分及抗氧化能力。結果顯示,野櫻莓於濃度2 mg/mL時DPPH自由基清除能力為74.89%;野櫻莓具有清除79.88%超氧陰離子活性;抑制黃嘌呤氧化酶活性為59.32%,顯示野櫻莓具有良好的抗氧化能力。在抗氧化成分分析方面,總酚含量為34.30 mg GAE/g dry weight,總類黃酮含量為5.49 mg QE/g dry weight,顯示野櫻莓具有豐富的抗氧化成分。野櫻莓中主要活性成分為綠原酸,其含量為4.45 mg/g dry weight。野櫻莓濃度5-20 mg/mL時,並無細胞毒性現象;胞內活性氧分子測定中,野櫻莓能使經Monosodium Urate Crystal(MSU)誘導之細胞其Reactive Oxygen Species(ROS)生成量降低,且於濃度20 mg/mL下與正對照組Allopurinol(100 μg/mL)之抑制ROS生成量無顯著差異,顯示野櫻莓可顯著抑制胞內活性氧的生成,並避免細胞受到氧化壓力之損傷。綜上所述,野櫻莓含有高含量的酚酸及類黃酮,具有良好的抗氧化活性,因清除超氧陰離子及抑制與尿酸生成有關之黃嘌呤氧化酶(抑制率59.32%),顯示具降尿酸之潛力,可開發相關機能性產品。 |

As the dietary patterns and lifestyles change, the incidence and prevalence of chronic diseases among our country's people are increasing yearly. Black chokeberry (Aronia melanocarpa) is rich in phenolic acids and flavonoids. These bioactive compounds have potential health benefits. This research aimed to investigate the bioactive compounds and antioxidant capacity of black chokeberry. The results showed that at a concentration of 2 mg/mL, the DPPH free radical scavenging ability of black chokeberry was 74.89%, and the scavenging capacity of superoxide anion was 79.88%. It also exhibited a xanthine oxidase inhibiting activity of 59.32%, the results indicated that black chokeberry possesses excellent antioxidant capacity. Regarding the analysis of antioxidant component, the total phenolic content was 34.30 mg GAE/g dry weight, and the total flavonoid content was 5.49 mg QE/g dry weight, showing that black chokeberry has a rich content of antioxidant component, and the primary active compound is chlorogenic acid (4.45 mg/g dry weight). In cell experiments, when the concentration of black chokeberry ranged from 5-20 mg/mL, it will not cause cytotoxicity. In the determination of intracellular reactive oxygen molecules, black chokeberry were able to reduce the reactive oxygen species (ROS) production induced by monosodium urate crystal (MSU) in cells. Moreover, there was no significant difference in the ROS production of the positive control group Allopurinol (100 μg/mL) at a concentration of 20 mg/mL, showing that black chokeberry can inhibit the production of intracellular reactive oxygen species to prevent cells from oxidative stress damage. In summary, black chokeberry contains high levels of phenolic acids and flavonoids, demonstrating excellent antioxidant activity. Black chokeberry has the potential to lower uric acid levels by scavenging superoxide anion activity and inhibiting xanthine oxidase (inhibition rate 59.32%) related to uric acid production. Therefore, it can be developed as a functional food products. |
