研究經由抗發炎活性篩選,發現『食茱萸莖部』之甲醇抽出物,對fMLP所誘導『超氧陰離子(O_2^(.-))』的產生與『彈性蛋白酶(elastase)』的釋放,具有顯著的抑制活性,顯示其具有研究開發的價值。取食茱萸莖部陰乾後,以MeOH冷浸抽取,所得到的MeOH抽取液經減壓濃縮後,再以EtOAc:H_2O進行分配,得到EtOAc可溶部,而H_2O層繼續以H_2O:n-BuOH進行分配,分別得到n-BuOH可溶部及H_2O可溶部。取EtOAc可溶部,利用大型管柱層析進行初步分離,再經由反覆的管柱層析、高效能液相層析(HPLC)及prep. TLC分離純化,共分離得到11種已知化合物。在這游離出的化合物中,呈現具有強而有力的抑制fMLP誘導人類嗜中性球產生超氧陰離子的能力,IC_(50) ≤ 5.46 μg/mL。

英文摘要

Through the screening of anti-inflammatory activity, it was found that the methanol extract of "Fructus Cornus officinale" significantly inhibited the production of "superoxide anion (O_2^(.-))" and the release of "elastase" induced by fMLP Activity, showing that it has the value of research and development. After drying the stems of Cornus officinalis in the shade, they were extracted by cold immersion in MeOH. The obtained MeOH extract was concentrated under reduced pressure, and then distributed with EtOAc:H_2O to obtain the EtOAc soluble part, while the H2O layer continued to be mixed with H_2O:n-BuOH Partitioning is carried out to obtain n-BuOH soluble part and H2O soluble part respectively. The soluble fraction of EtOAc was taken and used for primary separation by large-scale column chromatography, followed by repeated column chromatography, high performance liquid chromatography (HPLC) and prep. TLC separation and purification, and a total of 11 known compounds were isolated and obtained. Among the dissociated compounds, it exhibited a strong ability to inhibit fMLP-induced superoxide anion production in human neutrophils, IC_(50) ≤ 5.46 μg/mL.
