Purification requires expensive equipment and high operation cost for most methods to produce pure lipase. In this article, a simple defatting extraction method for large scale production of cheap lipase, RBL, from fresh rice bran was developed. The RBL obtained from this method did not lose any activity in the thermal stability test. For the storage stability test, the RBL was found to remain quite active after incubated at both 4℃ and 40℃ for a period of 40 days. The RBL also showed its superior durability during repeated batch hydrolysis. From the result of kinetic testing, the RBL showed a strong affinity toward the substrate by its higher Km value, and its low value of Ea also showed the RBL was less thermally sensitive. Finally, the RBL was proved to have the capability of catalyzing the hydrolyses of soybean oil and waste edible oil. |
