The value of at least 99.99% of power of exclusion was required according to the guideline of relationship test from Taiwan Association of Forensic Sciences. It is below the requirement based on the popular system of CODIS 15 STR loci for the case of single parent tested. More polymorphic markers were needed to increase the power of exclusion. This study screened 331 random individuals in Taiwan Han population using GlobalFiler^(TM) Express PCR Amplification Kit including CODIS 15 STR loci, 6 new STR loci (D1S1656, D2S441, D22S1045, D10S1248, D12S391, SE33), 1 Y-STR locus (DYS391), amelogenin gene and Y indel marker. All population data of STR loci were tested with no significant in Hardy- Weinberg equilibrium (p > 0.05) except D19S433. There are OL alleles observed in D1S1656 (allele 16.1 in 2 samples) and SE33 (28.1). In the test of concordance, genotype results were same with those of AmpFlSTR Identifiler Plus Kit. The matching probability, matching probability of the most popular genotype, probability of paternity exclusion in duo and trio case are 1.13E-25, 8.94E-20, 99.9997% and 99.99999988%. This study showed that the power of exclusion is able to meet the requirement of the above guideline with this system of 22 STR loci. |
