During late pregnancy, serum levels of progesterone, prostaglandin E2 (PGE2), and calcitonin (CT) increase, with these increases corresponding to the critical bone growth phase of the fetus. Although this indicates a potential interaction between thyroid C cells and CT secretion, the exact nature of this interaction remains unclear. In this study, we examined how PGE2 affects cell cycle proteins in TT thyroid C cells. Briefly, TT cells were exposed to various concentrations of PGE2 (50, 100, and 500 nM) and analyzed using immunofluorescence at 3, 24, and 72 h to track changes in cyclin levels during cell division. Our initial findings indicated that 50 nM PGE2 affected the expression of cyclins in TT cells, increasing the levels of cyclins E1 and A2 and decreasing the levels of cyclin D3 at 3 h. At 24 h, the levels of cyclins D1 and B1 increased, with a corresponding decrease occurring in the levels of cyclin E2. At 72 h, the levels of cyclins D1, D3, and E2 increased. Further increasing the dose of PGE2 resulted in additional changes in the levels of cyclins over time. These subtle yet distinct effects of PGE2 on cyclins D1, D3, E1, E2, A2, and B1 indicate a delicate balance exists in the regulation of the TT cell cycle, and therefore, it holds potential as a therapeutic target. In summary, PGE2 may accelerate the cell division cycle in TT cells. |
