本文主要為建立分析方法以同時檢測尿液中之13種運動禁藥-麻醉性止痛劑及其代謝物,包括:丁基原啡因(buprenorphine)及其代謝物norbuprenorphine、吩坦尼(fentanyl)及其代謝物 norfentanyl、羥二氫嗎啡(hydromorphone)、美沙冬(methadone)及其代謝物2-ethylidene-1,5-dimethyl-3,3-diphenylpyrrolidine、羥二氫可待因酮(oxycodone)及其代謝物noroxycodone、羥二氫嗎啡酮(oxymorphone)、速賜康(pentazocine)及配西汀(pethidine)及其代謝物 norpethidine。尿液檢體水解後使用單一固相萃取管柱進行萃取,收集混合沖提液,注入串聯式液相層析質譜儀(liquid chromatography / tandem mass spectrometry)以電噴灑游離法(electrospray ionization)之正電模式分析,並採用多重反應監控模式(multiple reaction monitoring)收集分析結果。所有藥物在相對滯留時間(CV%≦0.76)與特徵離子相對反應強度(CV%≦7.09)皆具有極佳的再現性。偵測極限為 2.5~50 ng/mL。所有藥物的偵測極限皆符合世界反運動禁藥組織(World Anti-Doping Agency)所規定的最低要求執行極限。 |

This paper presents a screening method for simultaneous detection 13 narcotics and their metabolites in urine. These substances included buprenorphine and its metabolite norbuprenorphine, fentanyl its metabolite norfentanyl, hydromorphone, methadone its metabolite 2-ethylidene-1, 5-dimethyl-3, 3-diphenylpyrrolidine, oxycodone its metabolite noroxycodone, oxymorphone, pentazocine, pethidine its metabolite norpethidine. Urine samples were hydrolyzed and extracted by single solid phase extraction cartridge. Eluent was collected and then assayed by LCMS/MS in positive ionization mode using an electrospray ionization and multiple reaction monitoring as the acquisition mode. All compound showed good reproducibility in terms of both the relative retention times (CV%≦0.76) and the relative abundances of the diagnostic transitions (CV%≦7.09). The limit of detection were in the range of 2.5~50 ng/mL, thus satisfying the minimum required performance limits set by the World Anti-Doping Agency. |
