本實驗探討源自黑鯛(Acanthopagrus schlegeli)脾臟細胞株BPS-4經熱休克處理後,分析其熱休克蛋白質的合成及其動力學,並探討其與細胞的耐熱性的關係。細胞先行熱處理(35℃/2小時; 37℃/2小時或40℃/30分鐘),立刻用含35S-methionine的培養液標幟,再進行聚丙烯胺膠體電泳和雙向電泳,經放射性自動顯影,知主要的熱休克蛋白為hsp87,70和33kD。在正常的培養溫度28℃下,此三種蛋白質也是常在的。熱休克蛋白的增加量,隨處理溫度之增加而增加。溫度需高至37℃以上時,hsp87和hsp70才明顯增多,而hsp33必須至40℃以上的溫度處理才大量增加。40℃/30分鐘的熱處理,hsp70和33的合成主要是在受熱後2小時內進行,而hsp87則延至4小時仍合成。35℃/2小時和37℃/2小時的預熱處理可誘導BPS-4細胞的熱耐性。 |
英文摘要 |
Cultured spleen cells (BPS-4 cells) of black porgy (Acanthopagrus schlegeli) were exposed to the elevated temperature. After heat treatment (35℃/2 hr; 37℃/2 hr or 40℃/30 min), cells were labeled in and methionine-free medium with 35 S-methionine immediately, and studied by one dimension SDS-PAGE and two dimension gel electrophoresis analysis. At 28 ℃, three major heat shock proteins (hsp 87, 70 and 33) presented in cells were found. At sublethal high temperature, these proteins increased dramatically with the elevation of temperature. However the pattern of their synthetic kinetics were different. The synthesis of hsp 87 and 70 was obviously enhanced at temperature above 37℃, hsp 33 was only enhanced above 40℃. Meanwhile, the thermotolerance in BPS-4 cells to be acquired by preheating at 35 ℃/2hr or 37 ℃/2hr was also observed. |
