Introduction: Thyroid cancer is leading endocrine malignancy and its diagnosis has been increasing in recent years. Prolonged exposure to TSH results in nodular growth of the thyroid tissue, thereby increasing the risk for thyroid cancer. The presence of any associations or synergistic effects between the BRAFV600E mutation and altered TSH levels in the progression of papillary thyroid carcinoma (PTC) remains uncertain. Therefore, this study aimed to evaluate the potential relation between the TSH and thyroid hormone levels with the clinicopathological features of PTC and the status of the BRAF^(V600E) mutation. Methods: Enzyme-linked immunosorbent assay was used to determine fT3, fT4, and TSH serum levels. DNA extractions from FFPE and fresh thyroid tumor tissues were extracted using a genomic DNA kit. An ABI 3730xl genetic analyzer was used for DNA sequencing. Result: In our study, 46 patients with thyroid cancer, a total of 9 (19.6%) cases with thyroid tumors were determined to have the BRAFV600E mutation. In this study, the mean serum levels of TSH, which are used to evaluate thyroid function, were determined to be 2.44±1.75 µIU/ml. The average levels of fT3 were 4.37±1.7 pg/ml, while fT4 levels were 15.5±4.47 pg/ml. In particular, there were no significant gender differences in these hormone levels. When assessing the hormonal changes and evaluating thyroid function in the participants, 78.26% (n=36) were found to have euthyroid, 17.39% (n=8) had hypothyroidism, and 4.35% (n=2) had hyperthyroidism. Mean serum levels of TSH and thyroid hormones were determined with no statistically significant differences between the groups with and without BRAF^(V600E) mutation. In conclusion, we did not find any correlation between BRAFV600E and levels of TSH, and thyroid hormones.
