研究經由抗發炎活性篩選,發現『台灣厚距花枝葉』之甲醇抽出物,對fMLP所誘導『超氧陰離子(O_2^(.-))』的產生與『彈性蛋白酶(elastase)』的釋放,具有顯著的抑制活性,顯示其具有研究開發的價值。取台灣厚距花枝葉陰乾後,以甲醇冷浸抽取,所得到的甲醇抽取液經減壓濃縮後,再以乙酸乙酯:水進行分配,得到乙酸乙酯可溶部,而水層繼續以水:正丁醇進行分配,分別得到正丁醇可溶部及水可溶部。取乙酸乙酯可溶部,利用大型管柱層析進行初步分離,再經由反覆的管柱層析、高效能液相層析(HPLC)及prep. TLC分離純化,共分離得到15種已知化合物。在這游離出的化合物中,其抗發炎活性試驗之結果顯示Betulinic acid及Oleanolic acid對於fMLP/CB所誘導超氧陰離子(O_2^(.-))的產生,具有顯著的抑制活性,其IC_(50)值分別為2.98 ± 1.81及0.72 ± 0.13 μM;Betulinic acid及Oleanolic acid對於fMLP/CB誘導彈性蛋白酶的釋放亦具有良好的抑制活性,其IC_(50)值分別為4.91 ± 0.21, 1.51 ± 0.12及10.2 ± 1.96 μM。 |

Through anti-inflammatory activity screening, the study found that the methanol extract of "branches and leaves of Pachycentria formosana" has a significant effect on the production of "superoxide anion (O2.-)" and the release of "elastase" induced by fMLP. Inhibitory activity shows its research and development value. After drying in the shade, the branches and leaves of the Taiwan flower were cold-soaked in methanol and extracted. The resulting methanol extract was concentrated under reduced pressure and then distributed with ethyl acetate:water to obtain the ethyl acetate soluble part, while the water layer continued to be Water: n-butanol is distributed to obtain n-butanol-soluble part and water-soluble part respectively. The ethyl acetate soluble part was taken and preliminary separated by large-scale column chromatography, and then separated and purified through repeated column chromatography, high-performance liquid chromatography (HPLC) and prep. TLC. A total of 15 known species were separated. compound.
