此研究經由抗發炎活性篩選,發現『黃槿莖部』之甲醇抽出物,對fMLP所誘導『超氧陰離子(O2.-)』的產生與『彈性蛋白酶(elastase)』的釋放,具有顯著的抑制活性,顯示其具有研究開發的價值。取黃槿莖部陰乾後,以MeOH冷浸抽取,所得到的MeOH抽取液經減壓濃縮後,再以CHCl3:H2O進行分配,得到CHCl3可溶部,而H2O層繼續以H2O:n-BuOH進行分配,分別得到n-BuOH可溶部及H2O可溶部。取CHCl3可溶部,利用大型管柱層析進行初步分離,再經由反覆的管柱層析、高效能液相層析(HPLC)及prep. TLC分離純化,共分離得到7種已知化合物。在單離出化合物中,Hibiscusamide(2)、N-trans-feruloyltyramine(3)及N-cis-feruloyltyramine(4)對於P-388(老鼠血癌細胞)產生均具有良好的細胞活性(IC50 < 4 μg/mL);另Hibiscusamide(2)對於HT-29(人類腸癌細胞)也具有良好的細胞活性(IC50 <4 μg/mL)。

英文摘要

This study screened for anti-inflammatory activity and found that the methanol extract from the stems of Hibiscus hibiscus has a significant effect on the production of "superoxide anion (O_2.-)" and the release of "elastase" induced by fMLP. Inhibitory activity shows its research and development value. After drying the Hibiscus stems in the shade, extract them by cold soaking with MeOH. After the MeOH extract is concentrated under reduced pressure, it is then distributed with CHCl_3:H2_O to obtain the CHCl_3 soluble part, while the H_2O layer continues to be divided into H2O:n-BuOH. Distribute to obtain n-BuOH soluble part and H_2O soluble part respectively. The soluble part of CHCl_3 was taken and initially separated by large-scale column chromatography, and then separated and purified through repeated column chromatography, high-performance liquid chromatography (HPLC) and prep. TLC. A total of 7 known compounds were isolated. Among the isolated compounds, Hibiscusamide (2), N-trans-feruloyltyramine (3) and N-cis-feruloyltyramine (4) all have good cell activity (IC_(50) < 4 μg) for the production of P-388 (mouse blood cancer cell) /mL); Hibiscusamide (2) also has good cell activity against HT-29 (human intestinal cancer cells) (IC_(50) < 4 μg/mL).
