中文摘要

To investigate T cell Vβ genes expression in patients with ankylosing spondylitis (AS), and whether there are any different expressions of peripheral blood T cell receptor (TCR) Vβ in AS patients and their first-degree relatives (FDR). Methods. Twenty-four families with thirty-six HLA-B27+ AS patients and their 48 FDR were divided into three groups: HLA-B27+ AS patients (n=36), their FDR who had B27 positive (B27+, n=24) and their FDR who had B27 negative (B27-, n=24). The peripheral blood T cell receptor Vβ gene was investigated using 10 different Vβ antibodies and the flowcytometric method. Results. The data revealed a significantly higher expression of TCR Vβ14 in B27+ AS patients and their B27+ FDR than in B27- FDR (p<0.05). Moreover, CD8+ T cells had significantly increased expressions of peripheral TCR Vß3, Vß9, Vß14, and Vß20 in AS than in their B27- FDR (p= 0.01, 0.03, 0.01, 0.03 respectively). Moderate/ severe disease activity had a higher expression of TCR Vβ13.6 compared to mild disease activity in AS patients. Conclusion. These results imply that TCR Vβ 14 chain usage was a preferred site for contact with the HLA-B27 molecule. AS patients had CD8+ T cells autoreactivity to a HLA-B27- restricted self-epitope. TCR Vβ 13.6 may be associated with the disease activity of AS.
